疾病概述 肿瘤(tumour)是指机体在各种致瘤因子作用下,局部组织细胞增生所形成的新生物(neogrowth),因为这种新生物多呈占位性块状突起,也称赘生物(neoplasm)。根据新生物的细胞特性及对机体的危害性程度,又将肿瘤分为良性肿瘤和恶性肿瘤两大类,而癌症即为恶性肿瘤的总称。
实验动物背景信息

p53基因剔除小鼠是一个自发肿瘤的模型小鼠,

C57BL/6J小鼠为近交系动物,毛为黑色。1975年我国从日本国立肿瘤研究所引进,引起对麻疹病毒敏感,对化学致癌剂不敏感,乳腺肿瘤发生率低,嗜酒精性高,干扰素产量高等特点,广泛用于免疫学、老年病学、肿瘤学、遗传学研究中。

模型制作方法

构建小鼠p53基因打靶载体(图1),p53引入一个无义突变,电转入ES细胞获得经基因修饰的打靶干细胞克隆,显微注射ES细胞到C57小鼠的受精卵中,移植到假孕小鼠,获得崁合体后代小鼠,再和C57小鼠杂交,最终获得可以传代的p53-/-小鼠(图2)。

模型表型数据

p53基因剔除小鼠在老年会发生多种肿瘤是一个自发肿瘤的模型小鼠,同时也是体内研究细胞生长、凋亡、放射损伤、寿命等的小鼠模型。

约74%的p53-/-小鼠6月龄前自发产生多种肿瘤,某些肿瘤甚至10周龄之前就已经出现。15-25周龄,肿瘤发生率迅速增加,肿瘤发生平均时间为20周龄。大部分肿瘤为恶性淋巴瘤,涉及器官多为胸腺和六大脏器如心、肺、脾、肝、肾、脑等(表1),成淋巴细胞细胞类型(图3)。
